Endocannabinoids in the Hedgehog pathway

We propose to identify the mechanisms by which endocannabinoids repress activity of the Hedgehog signaling pathway.   We will use clickable and photocrosslinkable endocannabinoid analogues to identify binding partners relevant to Hedgehog signaling, and to study the effects of pathway components on endocannabinoid trafficking.  Ultimately, reconstituting endocannabinoid trafficking and receptor binding in purified systems will lead us to a mechanistic understanding of their action.
